The aim of this project is to identify the key transcriptional circuitry that determines epithelial cell fates, specifically of epidermal and corneal limbal cells. As p63 is a key regulator of epidermal and corneal development and disease, this project takes a p63-centered approach to identify the transcription factors and their hierarchy in these stratified epithelia.
- Perform analyses of transcriptomics, epigenomics (ChIP-seq and ATAC-seq), proteomics in combination with computational modeling (supported by our computational experts)
- Use mature epidermal keratinocytes, corneal limbal/epithelial cells and the induced pluripotent stem cell (iPSC) differentiation as the cellular models for the multi-omics analyses
- Integrate these multi-omics data to identify the key transcription factors and their hierarchy Single-cell approaches such as single-cell RNA-seq may also be used in the project
- Have the opportunity to develop novel ideas and work on an independent project that aligns with our research interests
- A master degree in molecular or cellular biology, molecular life sciences or related field.
- Experience in in one or more of the following areas: molecular biology and cell biology techniques, generation and analysis of Next Generation Sequencing data, single cell approaches, (mammalian) genomics and gene regulation
- Experience in stem cell culture and/or computational analysis is preferred but not required
- Excellent communication skills in written and spoken English
- Strong analytical skills, a problem-solving and an result-oriented attitude
- The ability to work and interact with others in an energetic and supportive research environment
- Highly motivated and ambitious
The PhD student will join our research team in the Department of Molecular Developmental Biology, located at the Radboud Institute for Molecular Life Sciences (RIMLS), Faculty of Science in Nijmegen, the Netherlands. We offer a first class international research environment in regulation of gene expression and epigenetics using state-of-the-art genomic and proteomic technologies and bioinformatics. We run Next-Generation Sequencers, single cell equipment, Mass Spectrometers and accommodate a bioinformatics team and a local computational infrastructure.
